Applications of BBa_K3747140

We expressed the nos construct in Pseudomonas putida EM42 together with a plasmid containing possible electron donors for Nos (BBa_K3747141) to establish denitrification in this organism.

As the nos construct contains a rhamnose inducible promoter, the optimal rhamnose concentration (0.23 mM) by performing a growth experiment in which the OD600 was measured continuously over time (Figure 1).

P. putida EM42 containing pSEVA22-NosRZDFYLTatA and pSEVA83- ApbE-blue copper protein 1 – blue copper protein 2 – c550 (Nos). As control, P. putida EM42 containing pSEVAb22 without cargo was used (Control). Three biological replicates were used for the control and two biological replicates for Nos.
